Each champ has a unique formula for their signature, where you can plug in the signature level and get the result back. Below is the current list of "ideal" sig levels for each character.
For example, Abomination at level 20 has a 10.8% chance of poisoning.
The formula is = SM * log(sig, 2) + SB where SM is a multiplier and SB is a base. Each character's multiplier and base are different.
If you know that formula, you can take the derivative of it. I personally decided that when the derivative equals 0.5, using more sig stones becomes less efficient.

Yellow Jacket and Unstoppable Colossus have an ideal level of 1, meaning if you dupe them, that's all you need. Any extra levels are nice, but not really important. Hawkeye's signature is an example of a not good signature,requiring 99 levels for optimal performance.
